1. Results of cultivation in E. coli
Transformation of the BioBrick and cultivation of previously transformed E. coli resulted in a red bacterial culture. Tthe cell pellet of E. coli transformed with BBa_K593004 obtained via centrifugation was purple (see Fig. 1).

2. Sequence
We determined the nucleotide sequence of the BioBrick BBa_K593004 using sequencing techniques. Sequencing data showed that the BioBrick BBa_K593004 we received did not contain mmoX. The alignment of BBa_K593004 and BBa_K1390004 showed that the original BioBrick and our improved one differed substantially(see Fig.2). Blast analysis revealed that it actually contained parts of the Mini-Tn7 delivery vector pUC18T-mini-Tn7T-Zeo-DsRedExpress (Fig.3).

3. Improvement
The new part BBa_K1390004 now contains the original mmoX sequence from M. capsulatus (Bath), which was optimized for RFC10 Standard by removing restriction sites.
